On-chip Local Ph Measurement around Individual Cell Using Optically Manipulated Gel-tool with Adhesive-controllability

چکیده

A local pH measurement around cell by pH sensing gel-tool with adhesivecontrollability was developed. We fabricated gel-tool impregnated with pH indicator Bromothymol blue (BTB), which is used for pH measurement in neutral range. Adhesion of the gel-tool to objects such as glass, gel-tool and cell is performed by UV illumination in an electrolyte solution of a certain concentration. Gel-tool is made by salting-out of hydrophilic photo-crosslinkable resin ENT-3400 (Kansai Paint. Co., Ltd.). The pH sensing gel-tool is manipulated by optical tweezers. pH value is measured by detecting the color of the gel-tool. We succeeded in local pH measurement around a yeast cell by putting it into the ring-shaped gel-tool. Moreover, pH measurement of the cell was done by immobilizing it on the pH sensing gel-tool.
